Concept
The papanicolaous staining is the most used staining method for cytological material. First, the cores are stained with a haematoxylin solution. The cores appear blue, dark violet or black. The second step is the cytoplasm staining with an orange staining solution which shows especially the matureand cornified cells. The target structures are stained in orange of different intensities. In the third staining step, the polychrome solution is used. It is a mix of eosin, light green SF and Bismarck brown. The polychrome solutions shows the differentiation of the squamous epithelium.
Application
The polychrome solutions EA 31 and EA 50, papanicolaous solution 3a and 3b, are normally used for gynaecological materials. Polychrome solution EA 65, papnicolaous solution 3c and 3b are normally used for mucilaginous, non-gynaecological materials. Papanicolaous solution 3c shows a red staining, papanicolaous solution 3d a blue-green one.
