Permanent preparations are covered with covering agents which are puton the preparation in dissolved form at first and then harden by evaporation of the solvents.
Preparation:
Glass hard Canada balsam is dissolved in xylol for usage, viscous Canada balsam is diluted with xylol until it has the consistency of low viscosity honey.
Implementation:
Histological and cytological preparations must be completely dewatered before covering. As a last step, xylol or a xylol substitute should be used to avoid turbidities by aqueous solutions.
Covering takes place by dropping about 0.5 ml of a covering agent with a glass rod onto the slide in horizontal position to fill the space between slide andcover glass. When a homogeneous distribution over the preparation is ensured, carefully put a lean cover glass on it to avoid air bubbles. Then, the preparation stays in horizontal position until after about 20-30 min it is dry and can be microscoped.
The preparations treated this way are colour-stable for at least 5 years.
In the cytology, for the homogeneous coating of the preparations, Mercko glass is used instead of cover glasses. Apply a few drops to the preparation and ensure the uniform distribution of the covering agent. After the evaporation of the solvents, it comes to a hard, protective paint surface.
Entellan® New for covering machines is suited for common covering machines that work with glass coverings. The covering agent Entellan® New for covering machines is used according to the manual of the covering machine. The ideal amount of covering agent is determined at the first run on empty cover glasses and slides according to the cover glass size/pattern size/thickness and checked when the bottles are replaced.
